Currently testing the ability of PrP-res (prions) from mule deer and elk to convert PrP-sen from a variety of cell types, including humans, using the in vitro conversion assay to screen for susceptible species. Biological Aid - Lab of Jerry M. Keith, Ph.D. / Therapeutic and Vaccine Development Section / Oral Infection and Immunity Branch / National Institute of Dental and Craniofacial Research / National Institutes of Health, Bethesda, MD. 6/98-12/98 1. Cloned and attempted to express a repressor protein of virulence factor production from Bordetella pertussis in Escherichia coli for purification and in vitro characterization. 2. PCR mutagenized regulatory proteins of the B. pertussis infectious life cycle and transformed them into a highly virulent B. pertussis strain for aerosol survivability/induction studies. 3. Constructed gene fusions with an unstable variation of green fluorescent protein to determine if B. pertussis expresses certain genes when it enters eukaryotic cells. Solnick, M.D., Ph.D. / Division of Infectious and Immunologic Diseases / Department of Internal Medicine / UC Davis School of Medicine, Davis, CA. 6/96-6/98 1. Searched for the rpoS gene in H. pylori for characterization purposes. 2. Attempted to isolate and biochemically analyze a fresh H. pylori strain from biopsies obtained from endoscopy patients. 3. Attempted to isolate a mutant strain of H. pylori capable of growth under prolonged cultivation in vitro for characterization purposes. 4. Studied the phylogenetics of H. heilmannii by sequencing its 16SrRNA subunit from various mammalian stomach biopsies. Research Assistant III - Joy Melnikow, M.D., M.P.H. / Family Practice Unit / UC Davis Medical Center, Sacramento, CA. 9/95-4/96 1. Statistically analyzed the beneficial difference in terms of efficiency between the educated and non-educated patient.
